Shank3 is a scaffolding protein that is densely associated with postsynaptic glutamatergic synapses and is an important part of the postsynaptic density (PSD), a specialized protein-dense structure attached to the postsynaptic membrane. It acts as a master organizer of the PSD because of its ability to interact with multiple key synaptic components, including glutamate receptor complexes, anchoring proteins, and the actin cytoskeleton [ 7].
